Ecophysiology of three habitat-forming seaweeds across a depth and latitudinal gradient in southeastern Australia
Seasonal patterns in the in situ ecophysiology of the common habitat-forming seaweeds Ecklonia radiata, Phyllospora comosa, and Macrocystis pyrifera were investigated at different latitudes and depths in southeastern Australia. We used multiple performance indicators (photosynthetic characteristics, pigment content, chemical composition, stable isotopes, nucleic acids) to assess the ecophysiology of seaweeds near the northern and southern margins of their range, along a depth gradient (E. radiata only), over a two year period (September 2010 – August 2012).

- Statement
- Seaweeds were sampled quarterly in Tasmania, and every 6 months in NSW for 2 years (total of 8 sampling occasions for Tas and 4 for NSW). Ecklonia radiata was sampled at a depth of ~10 m and ~30 m, Phyllospora radiata was sampled at ~6 m, and Macrocystis pyrifera was sampled from the subsurface canopy (~0.5-1 m). Sampling consisted of PAM fluorometry measurements and collection of tissue samples for the following measurements: • Rapid Light Curves (light- and dark-adapted) using PAM fluorometry in situ - parameters are derived from fitted RLCs using the double exponential decay model of Platt et al (1980) • Pigment concentrations (chlorophyll a, c, and fucoxanthin) using HPLC and spectrophotometry) • Tissue chemistry (%C, %N, C:N, stable isotopes δ13C and δ15N) using a thermo gas chromatograph and isotope radio mass spectrometer • Nucleic acid characteristics (concentration of RNA and DNA, RNA:DNA) using a novel extraction technique developed specifically for brown macroalgae

- A blue LED diving-PAM (Waltz) was used for all fluorometry. A Thermo Gas Chromatograph coupled to a Finnigan Mat Delta S isotope radio mass spectrometer operating in continuous flow mode was used for all tissue chemistry measurements. Fucoxanthin was measured using a Waters Acquity H-series UPLC™ coupled to a Waters Acquity Photodiode Array Detector with an Ethylene Bridged Hybrid (BEH) C-18 column (2.1 x 100 mm x 1.7 micron particles). Chlorophyll a and c were measured using a Dynamica HALO RB-10 Spectrophotometer.
